Journal of Controlled Release Perspective highlights CloudSpec for targeted LNP analysis

An independent Perspective by Jens Bæk Simonsen and Philipp Nicolas Spahn in the Journal of Controlled Release discusses the challenges of developing targeted lipid nanoparticles (tLNPs), including the need for analytical methods to characterize these complex formulations.

These particles use surface ligands to direct therapeutic RNA toward specific cells. As RNA medicines advance, tLNPs are drawing interest for applications such as delivering mRNA to T cells inside the body, an approach now being evaluated in clinical trials for certain blood cancers and autoimmune diseases.

Developing these therapies requires analytical methods that can characterize complex formulations, including their targeting ligands. In discussing ligand measurement, the authors specifically mention Marama Labs’ CloudSpec:

“We suggest that UV-Vis-based approaches, such as CloudSpec's Scatter Free Absorbance from Marama Labs or similar methods used to measure mRNA directly in LNP samples, may also be capable of providing information on ligand concentration, provided that the absorption profiles of the mRNA and ligand-lipid conjugate can be distinguished or deconvolved.”

The authors note that this potential application depends on whether the absorption profiles of the mRNA and ligand-lipid conjugate can be distinguished or deconvolved. We’re pleased to see CloudSpec included in this independent discussion of how tLNPs can be better characterized.
